辣根过氧化物酶(HRP)是目前广为流行的一个标记酶。HRP成色反应方程为: HRP+H_2O_2←→[HRP.H_2O_2] (1) [HRP.H_2O_2]+(CHR)H_2←→(CHR)+ HRP+H_2O (2) 其中(CHR)H_2为供氢体,(CHR)为其氧化变色产物。测定HRP活力的方法有愈疮木酚法,联大茴香胺法,2、3′、6—三氯碘酚及二氨基联苯胺等方法。有些测定范围达1—25ng(30u/mgHRP)。但对于测定更微量的HRP,如欲分析某一很小神经组织内HRP的摄取和转运及在一定条件下的变化,以上这些方法的灵敏度尚嫌不足。因此需要摸索一种灵敏度更高的测定方法。据报道,目前HRP组化呈色反应常用的
