目的 探讨小牛血清去蛋白注射液对局灶性脑缺血大鼠脑血管化学储备能力的影响。方法将健康SD大鼠随机分为假手术组、单纯缺血组和小牛血清去蛋白注射液(Deproteinized calf blood serum injection,DCBSI)组。DCBSI组于术前3 d经大鼠腹腔注射DCBSI(2 ml/kg),每日1次,末次给药时间为术前5 h。复制大鼠局灶性脑缺血模型,并分别于术后3、6、24、48和72 h进行超氧化物歧化酶(Superoxide dismutase,SOD)活力及丙二醛(Malondialdehyde,MDA)含量的检测。结果与假手术组相比,单纯缺血组和DCBSI组大鼠脑组织匀浆SOD活力均明显降低(P<0.05),且随着缺血时间的延长而呈逐渐降低趋势,于48 h达最低;MDA含量均明显升高(P<0.05),且随着缺血时间的延长而呈逐渐升高趋势,于48 h达最高。与单纯缺血组相比,DCBSI组大鼠脑组织匀浆SOD活力均明显升高(P<0.05),MDA含量均明显降低(P<0.05)。结论小牛血清去蛋白注射液能够提高局灶性脑缺血大鼠脑血管的化学储备能力。
Objective To investigate the effect of deproteinised bovine serum on cerebral blood chemical reserve in focal cerebral ischemia rats. Methods Healthy Sprague-Dawley rats were randomly divided into sham operation group, ischemia group and Deproteinized calf blood serum injection (DCBSI) group. The rats in DCBSI group were intraperitoneally injected with DCBSI (2 ml / kg) once a day for 3 days before operation, and the last administration time was 5 hours before operation. The model of focal cerebral ischemia was established and the activities of superoxide dismutase (SOD) and malondialdehyde (MDA) were determined at 3, 6, 24, 48 and 72 h after operation Detection. Results Compared with the sham-operation group, the SOD activity in the homogenate of the ischemic group and DCBSI group was significantly decreased (P <0.05), and gradually decreased with the prolongation of ischemia time. At 48 h (P <0.05), and gradually increased with the prolongation of ischemia, reaching the highest at 48 h. Compared with the ischemia group, the SOD activity in the homogenate of DCBSI group was significantly increased (P <0.05) and MDA content was significantly decreased (P <0.05). Conclusion The deproteinization of bovine serum can improve the chemical reserve of cerebrovascular in focal cerebral ischemia rats.
