目的 :比较补阳还五汤及其有效部位组方对脑缺血后热休克蛋白 70 (HSP70 )表达的影响。方法 :采用沙土鼠双侧颈总动脉夹闭脑缺血模型 ,分别腹腔注射补阳还五汤及其有效部位组方 ,于缺血15min再灌注 4 8h后 ,以Northernblot检测脑组织HSP70mRNA的表达 ,以Westernblot检测其蛋白表达。结果 :沙土鼠脑缺血再灌注后 ,脑组织HSP70mRNA和蛋白表达显著增加 ,补阳还五汤及其有效部位组方可使脑缺血再灌注后HSP70mRNA表达减少 ,补阳还五汤的抑制作用强于有效部位组方 ;但两者对脑缺血再灌注后HSP70蛋白表达的增加均无明显影响。结论 :补阳还五汤及其有效部位组方可能通过抗缺血作用而抑制了缺血后HSP70mRNA的表达。
Objective: To compare the effect of Buyang Huanwu Decoction and its effective fraction on the expression of heat shock protein 70 (HSP70) after cerebral ischemia. METHODS: Cerebral ischemia model was established by bilateral common carotid artery occlusion in gerbils. The rats were sacrificed by intraperitoneal injection of Buyang Huanwu Decoction and its active fractions. The expression of HSP70 mRNA in brain tissue was detected by Northern blot after 48 min ischemia and 48 h after reperfusion. Western blot was used to detect its protein expression. RESULTS: After cerebral ischemia-reperfusion in gerbils, the expression of HSP70 mRNA and protein in the brain increased significantly. Buyang Huanwu Decoction and its active fractions can reduce the expression of HSP70 mRNA after cerebral ischemia-reperfusion, and inhibit the Yang Huanwu Decoction. The effect was stronger than that of the effective fraction; however, both had no significant effect on the increase of HSP70 protein expression after cerebral ischemia-reperfusion. Conclusion : Buyang Huanwu Decoction and its effective fractions may inhibit the expression of HSP70 mRNA after ischemia.
