目的:观察参附强心合剂对心衰大鼠神经内分泌系统、心肌组织病理学的影响。方法:选用SD雄性大鼠60只,采用腹主动脉缩窄法造心衰模型,造模成功的大鼠随机分为模型组、参附强心合剂大、小剂量组、西药组、假手术组,灌胃8周后,用放免法检测血浆和心肌血管紧张素II(AngII)含量,Elisa法检测血浆和心肌的去甲肾上腺素(NE)含量,并观察心肌组织的病理改变。结果:病理切片见,参附强心合剂大剂量组、西药组心肌细胞坏死及凋亡均显著下降;中药大剂量组及西药组与模型组相比能显著降低血浆和心肌AngⅡ、NE水平,有显著统计学差异(P<0.05)。结论:参附强心合剂可有效抑制心肌细胞坏死及凋亡,降低AngII、NE的水平,从而对缺血心肌有保护作用,并呈一定的量效关系。
Objective: To observe the effect of Shenfu Qiangxin Mixture on neuroendocrine system and myocardial histopathology in rats with heart failure. Methods: Sixty male Sprague Dawley rats were used to establish the model of heart failure by abdominal aorta constriction. The rats with successful model were randomly divided into model group, Shenfuxinxin large and small dose group, western medicine group, sham operation After 8 weeks, the content of angiotensin II (AngII) in plasma and myocardium was detected by radioimmunoassay. The content of norepinephrine (NE) in plasma and myocardium was detected by Elisa method and the pathological changes of myocardium were observed. Results: Pathological sections showed that Shenfuxin Mixture high dose group and Western medicine group significantly decreased myocardial necrosis and apoptosis; Compared with the model group, the Chinese medicine large dose group and the western medicine group could significantly decrease the levels of AngⅡ and NE in plasma and myocardium, There was a significant statistical difference (P <0.05). Conclusion: Shenfu Qiangxin Mixture can effectively inhibit the necrosis and apoptosis of cardiomyocytes, decrease the levels of AngII and NE, and thus have a protective effect on ischemic myocardium and show a certain dose-effect relationship.
