本文报道了31例(男性20例,女性11例;18~46岁)正常成人外周血淋巴细胞经X线体外照射后增殖能力的实验结果。标本采用全血和经葡聚糖-泛影钠密度梯度法分离的单核的细胞;照射剂量为0~400拉德;用集落形成和淋巴细胞刺激试验(LST)对照射细胞进行辐射敏感性分析。各实验组又分别用植物血凝素(PHA)、刀豆球蛋白A(Con-A)、A蛋白(PA)、美洲商陆(PWM)或结核菌素纯蛋白衍化物(PPD)作为促进细胞分裂的丝裂原,并用经2500拉德照射过的淋巴细胞作为刺激细胞,经0~400拉德照射后的淋巴细胞作为反应细胞,进行单向淋巴细胞混合培养(MLC)。分析不同方法所得的结果表明:用集落形成的方法检测淋巴细胞的辐射敏感性较LST更为灵敏,比较全血照射和分离的淋巴细胞照射后淋巴细胞的D_(37)值,发现全血照射在大部分的实验组中D_(37)值较相应淋巴细胞照射组为低。作者认为全血照射对淋巴细胞的损伤作用更大,其机理可能与密度梯度法分离的淋巴细胞中含更多的辐射抗性细胞有关,也不排除血内其他细胞的间接或直接影响。现将实验结果择要列表如下:
In this paper, we report the experimental results of proliferating ability of normal adult peripheral blood lymphocytes after X-ray irradiation in 31 cases (20 males, 11 females and 18-46 years old). The whole blood and mononuclear cells separated by dextran-sodium diacetate density gradient were used in the experiment. The irradiation dose was 0 to 400 rad. The radiosensitivity of irradiated cells by using colony formation and lymphocyte stimulation test (LST) analysis. The experimental groups were further boosted with PHA, Con-A, PA, PWM or PPD Cell division of mitogen, and irradiated with 2,500 rad irradiated lymphocytes as stimulating cells, 0 ~ 400 radiated lymphocytes as reactive cells, unidirectional lymphocyte mixed culture (MLC). Analysis of the results obtained by different methods showed that the radiosensitivity of lymphocytes detected by colony formation was more sensitive than that of LST. Comparing the D_ (37) values of lymphocytes after irradiation with whole blood and isolated lymphocytes, we found that whole blood irradiation D 37 in most experimental groups was lower than the corresponding lymphocyte group. The authors suggest that whole blood irradiation has a greater effect on lymphocyte injury, and its mechanism may be related to the more radiation-resistant cells in lymphocytes separated by density gradient method, and it also does not exclude the indirect or direct effects of other blood cells.
