目的观察培养人视网膜色素上皮细胞(RPE细胞)核因子κB(Nucleartranscriptionfactor-κBNF-κB)的表达,及地塞米松对其表达的影响。方法培养人RPE细胞接种于24孔板,随机分为阴性对照组、脂多糖(LipolysaccharideLPS)刺激组和地塞米松及LPS作用组,做免疫组化ABC法染色,显微镜下观察。结果对照组NF-κB在培养RPE细胞胞浆中表达,LPS刺激后转入胞核表达,地塞米松作用后,再以LPS刺激,RPE细胞核及细胞浆内NF-κB的表达减弱,胞核中表达水平减弱明显,且减弱的程度与地塞米松浓度有关。结论LPS能够激活RPE细胞的NF-κB使其向核内转入,一定浓度的地塞米松对RPE细胞的NF-κB表达及向胞核转入有抑制作用。
Objective To observe the expression of nuclear factor κB (NF-κB) in human retinal pigment epithelial cells (RPE) and the effect of dexamethasone on its expression. Methods Human RPE cells were seeded in 24-well plates and randomly divided into negative control group, lipopolysaccharide (LPS) -stimulated group, dexamethasone and LPS groups, and immunohistochemically stained with ABC method. Results The expression of NF-κB in the cytoplasm of RPE cells in the control group, the expression of NF-κB in the nucleus and cytoplasm of RPE cells after LPS stimulation and the expression of NF-κB in the cytoplasm of LEC stimulated by dexamethasone, In the expression level weakened significantly, and the degree of attenuation and dexamethasone concentration. CONCLUSION: LPS can activate NF-κB in RPE cells and transfer it into the nucleus. Dexamethasone can inhibit the expression of NF-κB and translocation into RPE cells.
