目的了解不同电压致兔坐骨神经电损伤后神经血流及Ⅰ型胶原的变化。方法将27只健康家兔电损伤后按损伤电压分为50、75、100 V组。分别于损伤后即刻(0)、1、4、8周,采用激光多普勒血流测定仪测定各组家兔坐骨神经血流量;免疫组织化学染色观察神经组织Ⅰ型胶原的变化情况,计算其阳性表达率。结果伤后0周,各组兔血流值均高于正常值,以后均降低。伤后8周,75、100 V组坐骨神经血流量[(53±3)、(48±5)灌注单位(PU)]均低于正常值[(62±4) PU,P<0.05]。50 V组胶原沉积不明显,75 V组及100 V组伤后4周神经外膜及束膜间可见较多的棕黄色阳性胶原纤维,伤后8周胶原纤维增生更为明显。伤后各组兔坐骨神经Ⅰ型胶原阳性表达率均高于正常值;75、100 V组伤后各时相点值均高于50 V组(P<0.01)。结论兔坐骨神经电损伤后神经血流的恢复随着损伤电压的增大而延缓,胶原纤维增生可能是神经血流减少的原因之一。
Objective To investigate the changes of neuroflood flow and type Ⅰ collagen after sciatic nerve injury induced by different voltage in rabbits. Methods 27 healthy rabbits were divided into 50, 75 and 100 V groups according to the lesion voltage after electric injury. Immediately after injury (0), 1, 4, and 8 weeks respectively, the blood flow of sciatic nerve in rabbits in each group was measured by laser Doppler flow cytometry. The changes of type Ⅰ collagen in nerve tissue were observed by immunohistochemical staining, Positive expression rate. Results At 0 week after injury, the blood flow of rabbits in each group were higher than the normal value and decreased later. At 8 weeks after injury, the sciatic nerve blood flow in the 75,100 V group was significantly lower than that in the normal group [(62 ± 4) PU, [(53 ± 3), (48 ± 5) PU) 50 V collagen deposition is not obvious, 75 V group and 100 V group 4 weeks after injury, there are more brown-yellow positive collagen fibers between the epineurium and the tunica intima, and collagen fibers hyperplasia is more obvious at 8 weeks after injury. The positive rate of type Ⅰ collagen in sciatic nerve in rabbits after injury was higher than that in normal group. The values of 75,100 V group were higher than those in 50 V group (P <0.01). Conclusion The recovery of nerve blood flow after sciatic nerve injury in rabbits is delayed with the increase of injury voltage. Collagen fiber proliferation may be one of the reasons of neuro-blood flow decrease.
