目的研究红木生育三烯酚(δ-T_3)对结肠癌小鼠CD4~+T细胞、CD8~+T细胞、白介素(IL)-6、干扰素(IFN)-γ的调节作用。方法 75只CD1-ICR小鼠分为结肠癌造模组和阴性对照两大组,结肠癌造模组再分为肿瘤模型对照组和三个δ-生育三烯酚剂量组(分别5、10、20mg/kg·bw),每组15只。采用氧化偶氮甲烷(AOM)和葡聚糖硫酸钠(DSS)诱导结肠癌模型。采用全自动五分类血球仪毛细管模式检测动物血液学指标,流式细胞仪检测外周血T淋巴细胞亚型,ELISA法检测血清白细胞介素(IL)-6、干扰素(IFN)-γ水平。结果与空白对照组比较,结肠癌造模组ICR小鼠白细胞计数降低(P<0.05)。与肿瘤模型对照组比较,中、高剂量生育三烯酚组均能显著提高CD4~+/CD8~+比值和血清中IFN-γ的水平,降低IL-6水平(P<0.05)。结论δ-生育三烯酚可增强结肠癌小鼠的免疫功能,这可能与其提高CD4~+/CD8~+比值,提高IFN-γ水平,及降低IL-6水平相关。
Objective To investigate the regulatory effect of matrine tocotrienol (δ-T_3) on CD4 ~ + T cells, CD8 ~ + T cells, interleukin (IL) -6 and interferon (IFN) -γ in colon cancer mice. Methods 75 CD1-ICR mice were divided into two groups: colon cancer model group and negative control group. The colon cancer model group was further divided into tumor model control group and three δ-tocotrienol dose groups , 20mg / kg · bw), 15 in each group. Colon cancer models were induced using azoxymethane (AOM) and dextran sodium sulfate (DSS). The hematological indexes of the animals were detected by capillary electrophoresis with five-category capillaries. The T-lymphocyte subsets in peripheral blood were detected by flow cytometry. The levels of serum interleukin-6 and interferon-gamma were measured by ELISA. Results Compared with the blank control group, the white blood cell count of ICR mice in colon cancer model group decreased (P <0.05). Compared with tumor model control group, the levels of CD4 ~ + / CD8 ~ + and serum IFN-γ and IL-6 levels in medium and high doses of tocotrienol increased significantly (P <0.05). Conclusion δ-Tocotrienol can enhance the immune function of colon cancer mice, which may be related to its increasing CD4 ~ + / CD8 ~ + ratio, increasing IFN-γ level and decreasing IL-6 level.
