目的探讨人单核细胞白血病细胞系SHI-1多药耐药谱及其机制。方法噻唑蓝(MTT)检测SHI-1细胞对柔红霉素、鬼臼乙叉甙、高三尖杉酯碱、紫杉醇和长春新碱的敏感性;流式细胞术(FCM)检测SHI-1细胞中P糖蛋白(P-gp)、多药耐药相关蛋白(MRP)、肺耐药相关蛋白(LRP)、乳腺癌耐药蛋白(BCRP)、谷胱甘肽-S-转移酶-π(GST-π)等耐药蛋白的表达。结果MTT药敏实验结果显示SHI-1细胞存在着多药耐药,尤其对VP-16最为明显。FCM检测显示SHI-1细胞中P-gp阳性比例10.78%,相对荧光强度(RFI)为1.02;GST-π阳性率93.52%,RFI6.91;LRP阳性率92.86%,RFI8.00;MRP阳性率4.54%,RFI1.38;BCRP阳性率3.84%,RFI1.06。结论SHI-1细胞存在多药耐药,以对VP-16最为明显,其耐药机制可能与LRP和GST-π的高表达有关。
Objective To investigate the multidrug resistance spectrum of human monocytic leukemia cell line SHI-1 and its mechanism. Methods Sensitivities of SHI-1 cells to daunorubicin, etoposide, homoharringtonine, paclitaxel and vincristine were determined by MTT assay. SHI-1 cells were detected by flow cytometry (FCM) P-gp, MRP, LRP, BCRP, Glutathione-S-transferase-π GST-π) and other resistant protein expression. Results The results of MTT assay showed that there were multidrug resistance in SHI-1 cells, especially VP-16. FCM showed that the positive rate of P-gp in SHI-1 cells was 10.78%, the relative fluorescence intensity (RFI) was 1.02, the positive rate of GST-π was 93.52%, the positive rate of RFI was 6.91, the positive rate of LRP was 92.86% 4.54%, RFI1.38; BCRP positive rate of 3.84%, RFI1.06. Conclusion SHI-1 cells have multi-drug resistance, which is most obvious to VP-16. The mechanism of drug resistance may be related to the high expression of LRP and GST-π.
