[目的 ]研究可见分光光度法在布鲁氏菌病试管凝集试验结果判定中应用。[方法 ]对布鲁氏菌病虎红平板试验中筛选出的4份样品(样品1、2为阳性,样品3、4为可疑),严格按《动物布鲁氏菌病诊断技术》(GB/T 18646—2002)中的试管凝集试验操作规程,对这4个奶牛样品的试管凝集试验结果进行人工判定和450/630nm分光光谱分析判读。[结果 ]人工判定样品1:100的结果为样品1、2、4可疑或阳性,样品3强阳性,分光光度法判读为样品1、2、4阳性,样品3强阳性。[结论 ]标准比浊管静置45 min后,各可见光谱数据均具有良好的线性关系;与人工判定结果相比,可见分光光度法判定的结果更直接、客观,受主客观影响因素更少。建议在标准GB/T 18646—2002的试管凝集试验操作规程中,增加可见分光光度法的判读标准。
[Objective] The research aimed to study the application of visible spectrophotometry in the judgment of brucellosis test tube agglutination test. [Method] Four samples (positive samples 1 and 2, samples 3 and 4) screened from brucellosis test were screened strictly according to the diagnostic criteria of brucellosis in animals (GB / T 18646-2002) in the tube agglutination test procedures, the four cow samples tube agglutination test results were manually determined and 450 / 630nm spectroscopic analysis read. [Result] The result of manual determination of sample 1: 100 was suspicious or positive of samples 1, 2 and 4, strongly positive of sample 3, the positive of samples 1, 2 and 4 was judged by spectrophotometry, and the sample 3 was strongly positive. [Conclusion] The standard turbidimetric tube was allowed to stand for 45 min, and each of the visible spectral data had a good linear relationship. Compared with the manual determination results, the results of the spectrophotometric determination showed that the results were more direct and objective and less affected by the subjective and objective factors . Proposed in the standard GB / T 18646-2002 test tube agglutination test procedures, increase the interpretation of visible spectrophotometry standards.
