目的:通过观测大鼠脾细胞中蛋白激酶C(PKC),B细胞淋巴瘤/白血病-2基因(Bcl-2)和Bax蛋白表达来探讨补脾方药对脾虚证的作用机制。方法:无特定病原体(SPF)大鼠采用复合方法制成脾虚模型后分别以3种不同配伍补脾方药灌胃3周。采用免疫组化法测定脾细胞中PKC,Bcl-2和Bax蛋白表达。结果:模型组与对照组比较,Bcl-2表达减少而Bax表达增加;中药组与模型组比较,Bcl-2表达增加而Bax表达减少;中药用药组与脾虚模型组比较,PKC表达减少(P<0.05),匀有统计学意义。结论:补脾方药治疗脾虚证的作用机制可能与提高脾细胞中Bcl-2表达,降低Bax和PKC表达有关。
Objective: To investigate the mechanism of invigorating the spleen recipe on spleen deficiency syndrome by observing protein kinase C (PKC), B cell lymphoma / leukemia-2 gene (Bcl-2) and Bax protein expression in rat spleen cells. Methods: SPF-free rats were made into spleen-deficiency model by compound method, and then were given 3 different prescriptions of Bu-spleen recipe for 3 weeks respectively. The protein expressions of PKC, Bcl-2 and Bax in spleen cells were detected by immunohistochemistry. Results: Compared with the control group, the expression of Bcl-2 and the expression of Bax increased in the model group and in the model group, while the expression of Bcl-2 and Bax in the model group decreased <0.05), with statistical significance. Conclusion: The mechanism of action of invigorating the spleen recipe on spleen deficiency syndrome may be related to increasing the expression of Bcl-2 and decreasing the expression of Bax and PKC in spleen cells.
