以1.6-dipheny1.3.5-hexatrience(DPH)作荧光探剂,应用荧光偏振技术测定30例Ⅱ型糖尿病(NIDDM)患者外周血中性粒细胞(polymorphonuclearleucocyte,PMN)膜脂流动性,并与24例健康人对照。结果显示,Ⅱ型糖尿病患者PMN膜脂流动性显著下降,膜微粘度明显增加,且膜脂流动性降低与空腹血糖水平和脂质过氧化程度呈负相关。提示PMN膜脂流动性降低可能是糖尿病病人白细胞功能异常易患感染的细胞结构基础,而长期高血糖状态和脂质过氧化增强则是导致糖尿病患者PMN膜脂流动性降低的重要因素。
Fluorescence polarization was used to determine the membrane fluidity of polymorphonuclear leucocyte (PMN) in 30 cases of type 2 diabetes mellitus (NIDDM) patients with 1.6-dipheny1.3.5-hexatrience (DPH) And with 24 healthy controls. The results showed that the fluidity of PMN membrane lipids decreased significantly in patients with type 2 diabetes mellitus, the micro-viscosity of the membrane increased significantly, and the decrease of membrane fluidity was negatively correlated with fasting blood glucose level and lipid peroxidation. It is suggested that the decrease of membrane fluidity of PMN may be the cellular structure basis of susceptible leucocyte dysfunction in diabetic patients. Long-term hyperglycemia and lipid peroxidation are the important factors leading to the decrease of PMN membrane fluidity in diabetic patients.
