目的观察苯甲酸雌二醇对人正常乳腺的影响。方法将9只裸鼠随机分为A、B、C3组,每组3只。取3例人正常乳腺组织标本,并将每例标本分为3份,分别植入各组中的1只裸鼠。植入2周后,从A组取出部分植入组织作对照组,之后A、B、C组裸鼠开始分别肌肉注射苯甲酸雌二醇3、6、9μg,每天1次,共28d;后处死裸鼠,取出植入组织。采用免疫组化方法检测各组植入组织的增殖细胞核抗原(proliferatingcellnuclearantigen,PCNA)、雌激素受体(estrogenreceptor,ER)、孕激素受体(progesteronereceptor,PR)的蛋白表达。结果与对照组相比,B、C组乳腺上皮细胞PCNA表达明显升高(P<0.05),A组乳腺PCNA无明显改变。随苯甲酸雌二醇剂量升高,ER表达逐渐下降(P<0.05),PR表达则逐渐上升(P<0.05)。结论一定剂量的苯甲酸雌二醇可促进正常乳腺的增生。
Objective To observe the effect of estradiol benzoate on human normal breast. Methods Nine nude mice were randomly divided into A, B and C3 groups, with 3 in each group. Three cases of normal breast tissue samples were taken, and each sample was divided into three, were implanted in each group of a nude mice. After 2 weeks of implantation, part of the implanted tissue was removed from group A as control group. Afterwards, nude mice in groups A, B and C began intramuscular injection of estradiol benzoate 3, 6 and 9 μg once daily for 28 days. Nude mice were sacrificed and the implanted tissue was removed. Immunohistochemistry was used to detect the protein expression of proliferating cell nuclear antigen (PCNA), estrogen receptor (ER) and progesterone receptor (PR) in each group. Results Compared with the control group, PCNA expression in mammary epithelial cells in groups B and C were significantly increased (P <0.05), but PCNA in group A was not significantly changed. With the increase of the dose of estradiol benzoate, the expression of ER gradually decreased (P <0.05) and the expression of PR increased gradually (P <0.05). Conclusion A certain dose of estradiol benzoate can promote normal breast hyperplasia.
