目的:研究黄芪水蛭制剂对糖尿病肾病大鼠IL-1β及C-Ⅳ、FN表达的影响,进而探讨其在糖尿病肾病大鼠肾保护中的作用机制。方法:采用高脂饲料喂养+STZ腹腔注射制作糖尿病肾病大鼠模型。分为正常对照组、模型对照组、中药治疗组(黄芪水蛭制剂组)、西药治疗组(洛汀新组)、雷公藤治疗组(雷公藤组)、中西药治疗组(黄芪水蛭制剂+洛汀新组),测定大鼠24 h尿蛋白定量、尿视黄醇结合蛋白、尿微量白蛋白,放射免疫方法测定大鼠血清中IL-β1及免疫组化方法测定大鼠肾脏组织中的C-Ⅳ、FN表达。结果:经黄芪水蛭制剂治疗2个月后,24 h尿蛋白定量、尿视黄醇结合蛋白、尿微量白蛋白、IL-1β各治疗组均明显低于模型组(P<0.05);大鼠模型组的C-Ⅳ、FN分别呈现高阳性表达,明显高于各治疗组(P<0.05)。结论:黄芪水蛭制剂对糖尿病肾病大鼠有一定的治疗效果,可以改善肾功能,延缓糖尿病的肾脏损伤,其可能机制是抑制了大鼠的炎症反应及肾纤维化的进展,与抑制了C-Ⅳ、FN在大鼠肾组织中的表达有关。
Objective: To study the effects of Astragalus leech on IL-1β, C-Ⅳ and FN expression in diabetic nephropathy rats and to explore its mechanism in renal protection of diabetic nephropathy rats. Methods: Diabetic nephropathy rats were induced by high fat diet and STZ intraperitoneal injection. They were divided into normal control group, model control group, TCM treatment group (Astragalus leech preparation group), Western medicine treatment group (Lotensin group), Tripterygium wilfordii group (Tripterygium wilfordii group), Western medicine treatment group (Astragalus leech preparation + Tingxin group). The levels of urinary protein, urine retinol binding protein, urine microalbumin and radioimmunoassay in 24 hours were measured to determine IL-β1 in rat serum and C -IV, FN expression. Results: The urinary protein excretion, urinary retinol binding protein, urine microalbumin and IL-1β in 24 h after treatment of Astragalus leech were both significantly lower than those in model group (P <0.05) C-Ⅳ and FN in model group showed high positive expression, which was significantly higher than that in each treatment group (P <0.05). Conclusion: Astragalus leeches preparation can improve renal function and delay kidney damage in diabetic nephropathy rats. Its possible mechanism is to inhibit the inflammatory reaction and the progress of renal fibrosis in rats, and to inhibit the effects of C- Ⅳ, FN in rat kidney tissue expression.
