目的:建立复方大黄软膏中黄芩苷的含量测定方法,并考察其体外透皮特性。方法:采用HPLC法测定黄芩苷含量,色谱柱为Agilent1200-C18柱(4.6 mm×250 mm,5μm),流动相为甲醇-水-磷酸(47∶53∶0.2),流速为1.0 m L/min,检测波长为280 nm;应用药物透皮扩散试验法考察其体外透皮特性。结果:黄芩苷的质量浓度在1.5172~242.75μg/m L范围内与峰面积呈良好线性关系(r=1),平均加样回收率102.1%(RSD%=2.7%,n=6);2批次复方大黄软膏中黄芩苷的含量分别为2.31 mg/g、2.17 mg/g,经离体裸小鼠皮肤24 h平均累积透过率仅为0.25%。结论:该方法简便、快速、准确,可用于复方大黄软膏的质量控制及透皮特性评价。
Objective: To establish a method for the determination of baicalin in compound rhubarb ointment and study its in vitro transdermal properties. Methods: The content of baicalin was determined by HPLC. The column was Agilent1200-C18 column (4.6 mm × 250 mm, 5 μm) with the mobile phase of methanol-water-phosphoric acid (47:53:0.2) , The detection wavelength was 280 nm; the drug transdermal diffusion test was used to investigate the in vitro transdermal properties. Results: The concentration of baicalin showed a good linear relationship with the peak area in the range of 1.5172 ~ 242.75μg / m L (r = 1), with an average recovery of 102.1% (RSD% = 2.7%, n = 6) The contents of baicalin in batch compound rhubarb ointment were 2.31 mg / g and 2.17 mg / g, respectively. The average cumulative transmission rate of the nude mouse skin after 24 h was only 0.25%. Conclusion: The method is simple, rapid and accurate and can be used for quality control and transdermal evaluation of compound rhubarb ointment.
