目的:建立同时测定茵栀黄分散片中绿原酸与栀子苷的含量测定方法。方法:采用HPLC法,Agilent ZorbaxSB-C18(色谱柱(250mm×4.6mm,5μm),流动相:乙腈-0.3%甲酸水,梯度洗脱,柱温30℃,流速:1.0mL.min-1,检测波长:0~9min为327nm(绿原酸),9~30min为238nm(栀子苷)。结果:绿原酸和栀子苷的线性范围分别为1.25~12.50μg.mL-1(r=0.999 6,n=6)、2.23~22.30μg.mL-1(r=0.999 8,n=6),平均回收率分别为99.82%(RSD为1.16%,n=6)与98.49%(RSD为2.27%,n=6)。结论:该方法简便、准确、重现性好,可用于茵栀黄分散片中绿原酸与栀子苷含量的同时测定。
Objective: To establish a method for simultaneous determination of chlorogenic acid and geniposide in Yinzhihuang dispersible tablets. Methods: The HPLC method was used on an Agilent Zorbax SB-C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-0.3% formic acid water and eluted with a gradient of 30 ℃. The flow rate was 1.0 mL · min- The detection wavelength was 327 nm (chlorogenic acid) at 0-9 min and 238 nm at 9-30 min.RESULTS: The linear ranges of chlorogenic acid and geniposide were 1.25-12.50 μg · mL -1 (r = The average recoveries were 99.82% (RSD 1.16%, n = 6) and 98.49% (RSD 0.999 6, n = 6) and 2.23-22.30μg.mL-1 2.27%, n = 6) .Conclusion: The method is simple, accurate and reproducible and can be used for the simultaneous determination of chlorogenic acid and geniposide in Yinzhihuang dispersible tablets.
