目的 :检测软骨细胞培养上清液中是否存在抑制血管生成的因子。方法 :用酶消化、分离鸡胚关节软骨细胞 ,体外单层培养 ,收集细胞培养上清液。并将该上清液作用于小鼠种植性实体瘤 S1 80 及培养人脐静脉内皮细胞 (HU VEC)上 ,探讨其抗血管形成作用的机制。结果 :软骨细胞培养上清液可抑制内皮细胞由 G1 期向 S期转换 ,使细胞停留于 G0 / G1 期。与对照组相比 ,增殖指数 (PI)明显下降 ,瘤重、肿瘤微血管密度 (intratumoral microvessel density,i MVD)也显著降低。结论 :软骨细胞培养上清液中具有抑制血管生成的活性因子
Objective: To detect whether there is a factor that inhibits angiogenesis in chondrocyte culture supernatant. Methods: Chondrocytes of chick embryo were digested by enzyme, cultured in monolayer in vitro, and the cell culture supernatants were collected. The supernatant was used to effect the anti-angiogenic effect of S180 on mouse xenograft solid tumor S180 and human umbilical vein endothelial cells (HU VEC). Results: The culture supernatant of chondrocytes could inhibit the transition from G1 phase to S phase and the cells stay in G0 / G1 phase. Compared with the control group, the proliferation index (PI) decreased significantly, tumor weight, tumor microvessel density (intratumoral microvessel density, i MVD) also significantly reduced. Conclusion: Chondrocyte culture supernatant has inhibitory activity of angiogenesis
