目的:通过对小半夏加茯苓汤高效液相色谱(HPLC)条件的考察,建立复方止呕效用成分,即鸟苷、盐酸麻黄碱、芦丁、6-姜酚、8-姜酚、10-姜酚、去氢茯苓酸及茯苓酸8种成分的特征图谱方法学。方法:分别考察复方50%醇提液HPLC分离的流动相、色谱柱、洗脱梯度及检测波长四方面因素。以RP-HPLC法梯度洗脱,Phenomenex Gemini C18色谱柱(5μ,250mm×4.6mm);乙腈-0.1%磷酸水溶液洗脱流动相;检测波长236nm;柱温40℃;流速1mL/min。结果:在一定范围内,鸟苷、盐酸麻黄碱、芦丁、6-姜酚、8-姜酚、10-姜酚、去氢茯苓酸及茯苓酸峰面积积分值Y与进样量X线性关系良好,加样回收率分别为98.33%、97.26%、97.81%、96.60%、98.01%、97.09%、96.46%和95.90%。结论:该方法操作简便、准确、重复性好,为复方质量标准评价提供了依据。
OBJECTIVE: To investigate the HPLC conditions of Xiao Ban Xia Jia Fu Ling Tang, to establish the compound anti-vomiting components, namely guanosine, ephedrine hydrochloride, rutin, 6-gingerol, Gingerol, Hydrogen Pericrin and Pachyrin acid Eight components of the characteristics of the mapping method. Methods: The mobile phase, chromatographic column, elution gradient and detection wavelength of 50% alcohol extract were investigated respectively. The mobile phase was eluted with a gradient of RP-HPLC on a Phenomenex Gemini C18 column (5μ, 250mm × 4.6mm); acetonitrile-0.1% phosphoric acid solution; the detection wavelength was at 236nm; the column temperature was 40 ℃ and the flow rate was 1mL / min. Results: In a certain range, the integral value of peak area of guanosine, ephedrine hydrochloride, rutin, 6-gingerol, 8-gingerol, 10-gingerol, The recoveries were 98.33%, 97.26%, 97.81%, 96.60%, 98.01%, 97.09%, 96.46% and 95.90%, respectively. Conclusion: The method is simple, accurate, reproducible and provides the basis for the evaluation of compound quality standards.
