目的 :建立能够模拟在体血脑屏障的体外模型。方法 :培养大鼠脑微血管内皮细胞和星形胶质细胞 ,用免疫组织化学染色的方法鉴定细胞 ,分别接种两种细胞于多聚酯多孔膜的两面 ,相差显微镜及HE染色观察细胞在多聚酯膜上的生长情况。结果 :培养的脑微血管内皮细胞多数为梭形铺路石样外观 ,而星形胶质细胞呈具有多个突起的典型形态 ,免疫组织化学染色鉴定细胞的纯度分别在 90 %和 95 %以上 ,两种细胞能够以单层细胞的形式生长于多聚酯多孔膜的两面。结论 :通过脑微血管内皮细胞和星形胶质细胞共同培养能够形成与在体血脑屏障类似的结构 ,是一种建立血脑屏障模型的可行方法。
Objective: To establish an in vitro model that can simulate in-vivo blood-brain barrier. Methods: The cultured rat brain microvascular endothelial cells and astrocytes were identified by immunohistochemical staining. Two kinds of cells were inoculated on both sides of the porous polyester membrane. The cells were observed under phase contrast microscope and HE staining. Ester film on the growth. RESULTS: The majority of cultured brain microvascular endothelial cells were spindle-shaped paving stone-like appearances, whereas astrocytes showed typical morphologies with multiple protuberances. The purity of the cells identified by immunohistochemical staining was above 90% and 95% The seed cells can grow on both sides of the multi-layer porous polyester film in the form of a monolayer of cells. CONCLUSION: Co-culture of brain microvascular endothelial cells and astrocytes is capable of forming a structure similar to that of the blood-brain barrier in the body and is a viable method for establishing a model of the blood-brain barrier.
