目的观察大黄素对角朊细胞体外增殖分化的影响。方法以人转化型角朊细胞株COLO-16细胞为模型,采用活细胞计数、3H-TdR参入法、细胞因子生物活性检测、电镜及ABC法进行研究。结果大黄素能以一种剂量依赖方式抑制角朊细胞的体外增殖和DNA合成;减少角朊细胞肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的产生;对角朊细胞超微结构有直接损伤作用;并抑制角朊细胞角蛋白表达,破坏细胞骨架。结论大黄素通过不同途径抑制角朊细胞的体外增殖分化,是角朊细胞增殖分化的有效抑制剂。
Objective To observe the effects of emodin on proliferation and differentiation of keratinocytes in vitro. Methods The human transformed keratinocyte cell line COLO-16 was used as a model, and the viable cell count, 3H-TdR incorporation assay, cytokine bioactivity assay, electron microscopy, and ABC method were used for the study. Results Emodin could inhibit the proliferation and DNA synthesis of keratinocytes in vitro in a dose-dependent manner, and reduce the production of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in keratinocytes. The ultrastructure of keratinocytes has a direct damaging effect and inhibits keratinocyte cytokeratin expression and disrupts the cytoskeleton. Conclusion Emodin inhibits the proliferation and differentiation of keratinocytes in vitro through different pathways and is an effective inhibitor of keratinocyte proliferation and differentiation.
