目的 研究 Na As O2 和 Na3As O4对 HL- 6 0细胞生长及基因组 DNA甲基化的影响。方法 采用细胞培养和限制性内切酶解分析技术观察分析细胞生长及细胞基因组 DNA甲基化水平。结果 1两种无机砷化物对 HL- 6 0细胞生长具有抑制作用 ,特别是高浓度 Na As O2 (2、10 μmol/ L)和 Na3As O4(10 0、30 0 μmol/ L)可明显地抑制 HL- 6 0细胞的生长 (P <0 .0 1)。2随着砷化物浓度的增加 ,Msp 酶解基因组 DNA片段的分子量变化不大 ,但 Hpa 酶解片段的分子量逐渐变大。结论 提示基因组 DNA中 CCGG核苷酸内侧 C的甲基化总的水平有上升趋势。
Objective To study the effects of Na As O2 and Na3As O4 on the growth and DNA methylation of HL-60 cells. Methods Cell culture and restriction endonuclease analysis were used to observe the cell growth and methylation of genomic DNA. Results 1 Two kinds of inorganic arsenic compounds could inhibit the growth of HL-60 cells. In particular, high concentrations of Na As O2 (2,10 μmol / L) and Na3As O4 (10 0,30 0 μmol / L) Growth of HL-60 cells (P <0.01). 2 As the concentration of arsenic increased, the molecular weight of Msp enzymatic genomic DNA did not change much, but the molecular weight of Hpa enzymatic hydrolysis became larger. The results suggest that the overall level of C methylation of CCGG nucleotides in genomic DNA is on the rise.
