目的:观察核糖核酸(RNA)对日本血吸虫病病兔肝纤维化的阻抑效果。方法:54只3月龄家兔随机均分为正常组、感染组及RNA组。于肌注RNA后第40d、70d及100d,各组随机选6只兔抽血及取肝组织,通过光镜、电镜观察其组织病理变化,放射免疫法测定血浆HA含量,紫外分光光度法测定肝组织HYP,IBAS图像分析仪测定肝组织胶原纤维分布面积百分比,用潘孺孙法测定肝组织胶原纤维含量。结果:与感染组相比较,RNA组胶原纤维未见明显增加,而肝细胞核仁增大,粗面内质网及其附着颗粒增多,贮脂细胞周围胶原纤维减少,血浆HA、肝组织HYP、胶原纤维分布面积百分比及胶原纤维含量均显著降低。结论:RNA对日本血吸虫病病兔肝纤维化具有早期预防作用。
Objective: To observe the inhibitory effect of ribonucleic acid (RNA) on hepatic fibrosis in rabbits with schistosomiasis japonica. Methods: 54 three-month-old rabbits were randomly divided into normal group, infection group and RNA group. At the 40th, 70th and 100th day after intramuscular injection of RNA, 6 rabbits in each group were randomly selected to take blood and liver tissues. The histopathological changes were observed by light microscope and electron microscope. The contents of plasma HA were measured by radioimmunoassay. The UV spectrophotometry Liver tissue HYP, IBAS image analyzer to determine the percentage of liver fibrosis area distribution of collagen, liver fiber collagen content was determined by Poon Sun method. Results: Compared with the infected group, there was no obvious increase in the collagen fibers in the RNA group, while the nucleolus of the hepatocytes increased, the rough endoplasmic reticulum and its attached granules increased, the collagen fibers around the fat storage cells decreased, the contents of HA, Collagen fiber distribution area percentage and collagen fiber content were significantly reduced. Conclusion: RNA has an early preventive effect on hepatic fibrosis in rabbits with schistosomiasis japonica.
