目的:通过对酸枣仁汤含药脑脊液指纹图谱的研究,为酸枣仁汤镇静催眠药效物质基础研究提供方法学基础。方法:采用反相高效液相色谱法,以梯度洗脱的方式,50 min内在同一图谱中同时完成水溶性成分和脂溶性成分指纹图谱的建立,同时将含药脑脊液、空白脑脊液、酸枣仁汤及各单味药HPLC指纹图谱进行对比分析。结果:通过对脑脊液采集方法、前处理方法及洗脱梯度、检测波长等色谱条件的筛选,建立了可将酸枣仁汤含药脑脊液中化学成分完全分开的指纹图谱,并进行归属分析,初步鉴定出含药脑脊液中的固有成分、药源性物质及代谢产物。结论:方法可作为探索酸枣仁汤药效物质基础研究的有效手段。
OBJECTIVE: To provide a methodological basis for the study of the sedative-hypnotic effect substances of Suanzaoren Tang by studying the cerebrospinal fluid fingerprint of Suanzaoren Decoction. Methods: The fingerprint of water-soluble and fat-soluble components was established simultaneously in 50 min by reversed-phase high-performance liquid chromatography with gradient elution. Meanwhile, the drug-containing cerebrospinal fluid, blank cerebrospinal fluid, And the single drug HPLC fingerprint comparative analysis. Results: The fingerprints which can separate the chemical components in the cerebrospinal fluid of the Suanzaoren Decoction drug solution were established by screening the methods of cerebrospinal fluid collection, pretreatment, chromatographic conditions such as elution gradient and detection wavelength. The drug-containing cerebrospinal fluid of the inherent components, drug-derived substances and metabolites. Conclusion: The method can be used as an effective means to explore the pharmacological basis of Suanzaoren decoction.
