目的:建立高效液相色谱法同时测定决明子药材中决明子苷B2、红镰霉素龙胆二糖苷及决明子苷C三个萘并吡喃酮成分的含量测定方法。方法:采用Sunfire C18色谱柱(250mm×4.6mm,5μm),柱温:35℃,以甲醇-乙腈(2∶1)混合溶液(A)-水(B)梯度洗脱,流速为1.0m L/min,检测波长为278nm。结果:决明子苷B2、红镰霉素龙胆二糖苷及决明子苷C分别在0.004335-0.5352、0.006444-0.7956、0.005657-0.6984μg,呈良好线性关系,平均回收率(n=6)分别为100.37%、98.89%和102.38%。含量测定结果显示不同来源的决明子饮片中3个成分含量差异较大。结论:本方法经方法学验证,可用于决明子药材的质量控制和评价。
OBJECTIVE: To establish a HPLC method for simultaneous determination of three naphthopyranones in cassia seed B2, erythromycin gentiobioside and cassia. Methods: A Sunfire C18 column (250 mm × 4.6 mm, 5 μm) was used. The column temperature was 35 ℃ and the gradient of methanol-acetonitrile (2:1) mixed solution (A) / min, detection wavelength of 278nm. Results: There was a good linear relationship between cassia seed B2, erythromycyxin gentiobioside and cassiacin C in the range of 0.004335-0.5352, 0.006444-0.7956 and 0.005657-0.6984μg, respectively. The average recoveries (n = 6) were 100.37% , 98.89% and 102.38% respectively. Content determination results show that different sources of cassia seed in three components of the larger differences. Conclusion: This method is validated by methodology and can be used for quality control and evaluation of cassia seed.
