目的观察丙种球蛋白对川崎病患儿血清诱导人单核细胞表达血小板源生长因子(PDGF)B链蛋白(PDGFB)的影响;观察丙种球蛋白作用后的单核细胞条件培养基(MCM)对内皮细胞凋亡的影响。方法应用免疫细胞化学和流式细胞术方法。结果川崎病患儿血清能强烈诱导单核细胞表达PDGFB,川崎病患儿血清作用后的MCM则能显著促进内皮细胞凋亡。川崎病患儿血清的上述作用能够被丙种球蛋白和丙种球蛋白作用后的MCM所抑制。结论单核细胞表达PDGF增多及单核细胞介导的内皮细胞凋亡,可能参与了川崎病患儿冠状动脉并发症的形成;丙种球蛋白防治川崎病患儿冠状动脉并发症的机理,可能与抑制单核细胞表达PDGF和减少内皮细胞凋亡有关
Objective To observe the effect of gamma globulin on the expression of PDGF-B in serum induced by Kawasaki disease in children with Kawasaki disease. The effects of gamma globulin on monocyte conditioned medium (MCM) ) On endothelial cell apoptosis. Methods Immunocytochemistry and flow cytometry were used. Results Kawasaki disease serum can strongly induce monocyte expression of PDGF B, Kawasaki disease in children with serum MCM can significantly promote endothelial cell apoptosis. The above-mentioned effects of Kawasaki disease serum can be suppressed by MCM after gammaglobulin and gammaglobulin. Conclusion The monocyte-derived PDGF expression and monocyte-mediated endothelial cell apoptosis may be involved in the formation of coronary artery disease in children with Kawasaki disease. The mechanism of prevention and treatment of coronary artery complications in children with Kawasaki disease may be related to Inhibition of monocytes expressing PDGF and reducing endothelial cell apoptosis
