目的 :探讨补肾方、活血方、补肾活血方对血管性痴呆小鼠学习记忆能力及血浆血栓素B2(TXB2)和6-酮-前列腺素1α(6-Keto-PGF1α)含量的影响。方法:采用反复脑缺血再灌注法复制血管性痴呆小鼠模型,以尼莫地平为对照组,观察补肾方、活血方、补肾活血方三组小鼠水迷宫法行为学及血浆TXB2和6-Keto-PGF1α含量的变化。结果 :与假手术组比较,模型组小鼠学习与记忆能力明显下降(P<0.01),血浆TXB2含量明显升高,6-Keto-PGF1α含量明显下降(P<0.01);与模型组比较,各治疗组学习记忆成绩及血浆6-Keto-PGF1α含量均明显提高,血浆TXB2含量明显降低(P<0.05或P<0.01);各治疗组比较,补肾活血组(生药14.28g/kg)的学习和记忆成绩优于其它治疗组(P<0.05),补肾活血组和活血组(生药7.14g/kg)在降低血浆TXB2,升高6-Keto-PGF1α含量优于补肾组(生药7.14g/kg,P<0.05或P<0.01)。结论:补肾方、活血方、补肾活血方均可通过降低血浆TXB2,升高6-Keto-PGF1α含量,改善血管性痴呆小鼠学习与记忆能力,且补肾活血方与活血方在调节TXB2/6-Keto-PGF1α作用方面优于补肾方。
Objective: To investigate the effects of Bushen Fang, Huoxue Fang and Bushen Huoxue Recipe on the learning and memory abilities and the content of TXB2 and 6-Keto-PGF1α in mice with vascular dementia. Methods: The model of vascular dementia was duplicated by repeated cerebral ischemia-reperfusion in rats. Nimodipine was used as the control group to observe the water maze behavior and the levels of plasma TXB2 and 6 in mice of Bushen Fang, Huoxue Fang and Bushen Huoxue Recipe -Keto-PGF1α content changes. Results: Compared with the sham-operation group, the learning and memory abilities of the model group decreased significantly (P <0.01), the content of TXB2 in the model group increased significantly and the content of 6-Keto-PGF1α significantly decreased (P <0.01) The levels of learning and memory and plasma 6-Keto-PGF1α in each treatment group were significantly increased, and the content of TXB2 in plasma was significantly decreased (P <0.05 or P <0.01). Compared with the treatment group, the study of Bushen Huoxue group (14.28g / kg crude drug) (P <0.05). Bushen Huoxue group and Huoxue group (crude drug 7.14g / kg) had lower TXB2 and higher 6-Keto-PGF1alpha than those in Bushen group (7.14g / kg , P <0.05 or P <0.01). Conclusion: Bushenfanghuoxue recipe and Bushenhuoxue recipe can both improve the learning and memory abilities of mice with vascular dementia by decreasing plasma TXB2 and increasing 6-Keto-PGF1α, and the effect of Bushen Huoxue recipe and Huoxue recipe in regulating TXB2 / 6 -Keto-PGF1α effect than Kidney side.
