目的:探讨舒心饮预给药对急性心肌缺血大鼠心肌细胞凋亡抑制因子(Fas)的影响,为舒心饮的临床应用提供依据。方法:将40只Wistar健康雄性大鼠随机分为对照组、模型组、舒心饮组、单硝酸异山梨酯组。对照组、模型组给予灌胃蒸馏水,舒心饮组灌胃2.43 g/kg生药,单硝酸异山梨酯组灌胃1 mg/kg。用药5周后,结扎左冠状动脉前降支造模,观察大鼠血清肌酸磷酸激酸(CPK)及乳酸脱氢酶(LDH)浓度,RT-PCR检测心肌肿瘤坏死因子-α(TNF-α)、Fas mRNA,Western blot检测心肌Fas、半胱氨酸蛋白酶-3(Caspase-3)表达水平,评价舒心饮对大鼠急性心肌缺血的作用。结果:舒心饮可明显降低血CPK、LDH浓度,下调TNF-α、Fas转录水平及Fas、Caspase3蛋白水平的表达。结论:舒心饮预处理对大鼠急性缺血心肌有保护作用,下调死亡途径的受体和配体从而拮抗心肌组织的凋亡可能是其作用机制之一。
Objective: To investigate the effect of Shuxin drink preconditioning on myocardial apoptosis inhibitor (Fas) in acute myocardial ischemia rats, and to provide basis for its clinical application. Methods: Forty Wistar healthy male rats were randomly divided into control group, model group, Shu Xin Yin group and isosorbide mononitrate group. In the control group and model group, intragastric administration of distilled water and Shuxin drink group were given 2.43 g / kg crude drug, and isosorbide mononitrate group was given intragastrically 1 mg / kg. After 5 weeks of treatment, the anterior descending artery of the left coronary artery was ligated and the concentrations of serum creatine phosphokate (CPK) and lactate dehydrogenase (LDH) were measured. The levels of tumor necrosis factor-α (TNF- α, Fas mRNA and Western blot were used to detect the expression of Fas and Caspase-3 in myocardium and the effect of Shu Xin Yin on acute myocardial ischemia in rats. Results: Shuxin Decoction can significantly reduce the concentrations of CPK and LDH in blood, down-regulate the expression of TNF-α, Fas and Fas, Caspase3 protein levels. Conclusion: Shuxin Decoction preconditioning has the protective effect on acute ischemic myocardium in rats. Down-regulating the receptors and ligands of death pathway to antagonize the apoptosis of myocardial tissue may be one of its mechanisms.
