目的:研究保妇康对慢性盆腔炎大鼠子宫粘膜细胞间黏附分子1(ICMI-1)和转化生长因子β1((TGF-β1)mRNA表达及血清炎性细胞因子含量的影响,以探讨其作用机制。方法:将雌性大鼠随机分为7组,除正常组和假手术组外,其他各组采用混合细菌加机械损伤法建立慢性盆腔炎大鼠模型。保妇康高、中、低剂量组分别以2.07,4.14,8.28g/kg ig给药,连续给药4周,采用半定量荧光免疫技术(RT-PCR)法测定大鼠子宫组织ICMI-1和TGF-β1mRNA的表达,用ELASA法测定血清TNF-α和IL-6的含量。结果:与正常组相比,模型组大鼠子宫黏膜炎性细胞浸润明显,子宫组织ICMI-1(2.03±2.76)和TGF-β1(9.03±2.76)mRNA的表达均明显升高(P<0.05)。与模型组比较,保妇康2.07~8.28g/kg剂量范围内,均可明显减轻子宫粘膜炎症反应,高剂量组(8.28g/kg)、中剂量组(4.14g/kg)ICMI-1和TGF-β1mRNA的表达均明显降低,其中,高剂量组TGF-β1mRNA(4.11±1.29)的表达下降最明显;低剂量组(2.07g/kg)ICMI-1和TGF-β1mRNA的降低没有显著性差异。高、中剂量组(4.14~8.28g/kg)的IL-6和TNF-α明显降低。结论:保妇康能降低血清炎性细胞因子的含量,下调子宫组织ICMI-1和TGF-β1mRNA的表达,减轻盆腔组织粘连,这可能是其防治慢性盆腔炎的机制之一。
Objective: To study the effect of Baofukang on the expression of ICMI-1 and TGF-β1 mRNA and serum inflammatory cytokines in rats with chronic pelvic inflammatory disease Methods: The female rats were randomly divided into 7 groups, except the normal group and the sham operation group, the other groups were treated with mixed bacteria plus mechanical injury to establish the model of chronic pelvic inflammatory disease.Pao Kang health high, medium and low Dose group were given ig at 2.07,4.14,8.28g / kg, continuous administration for 4 weeks, using semi-quantitative fluorescent immunostaining (RT-PCR) method to determine the expression of ICMI-1 and TGF-β1 mRNA in rat uterine tissue, ELASA method was used to detect the levels of TNF-α and IL-6 in serum.Results: Compared with the normal group, the infiltration of inflammatory cells in uterine mucosa of model group was obvious, the ICMI-1 (2.03 ± 2.76) and TGF-β1 ± 2.76) (P <0.05) .Compared with the model group, the bFGF significantly reduced the inflammation of uterine mucosa in the dose range of 2.07-8.28g / kg, while in the high dose group (8.28g / kg), and the expression of ICMI-1 and TGF-β1mRNA in middle-dose group (4.14g / kg) were significantly decreased. Among them, the expression of TGF-β1mRNA (4.11 ± 1.29) (P <0.01), and the most obvious decrease was noticed in the low dose group (2.07g / kg), while the levels of IL-6 and TNF-α in high and middle dose groups (4.14-8.28g / kg) .Conclusion: Baofukang can reduce serum levels of inflammatory cytokines and down-regulate the expressions of ICMI-1 and TGF-β1 mRNA in the uterus and reduce pelvic tissue adhesion, which may be one of the mechanisms of prevention and treatment of chronic pelvic inflammatory disease.
