根据先前获得的EST序列,从巴西橡胶树(Hevea brasiliensis)中克隆了一个编码磷酸烯醇式丙酮酸羧化酶激酶相关激酶(phosphoenolpyruvate carboxylase kinase-related kinase,PEPRK)的基因HbPEPRK1。HbPEPRK1编码区c DNA长度为1 401 bp,编码466个氨基酸。HbPEPRK1蛋白具有典型的的丝/苏蛋白激酶结构特征的结构特征,和其他植物中的类似蛋白具有很高的同源性。对HbPEPRK1启动子区域进行分析,发现含有众多应答激素和环境胁迫信号元件。实时荧光定量分析结果表明,HbPEPRK1在所检测的组织中均有表达,其中在胶乳的表达量显著高于其他组织;乙烯刺激后HbPEPRK1在胶乳中的表达量明显增加。
Based on the EST sequences previously obtained, a gene encoding HEPEP1 (HEPEP1) encoding phosphoenolpyruvate carboxylase kinase-related kinase (PEPRK) was cloned from Hevea brasiliensis. The length of c DNA of HbPEPRK1 coding region was 1 401 bp, encoding 466 amino acids. The HbPEPRK1 protein has typical structural features of filament / thuin kinase structural features and has high homology with similar proteins in other plants. The HbPEPRK1 promoter region was analyzed and found to contain numerous response hormones and environmental stress signaling elements. The results of real-time fluorescence quantitative analysis showed that HbPEPRK1 was expressed in the tested tissues, and the expression level of HbPEPRK1 in latex was significantly higher than that in other tissues. The expression level of HbPEPRK1 in latex was significantly increased after ethylene stimulation.
