目的 :探讨抗纤方对TGF β1诱导肝细胞凋亡的抑制作用。方法 :采用细胞生物学等方法 ,利用TGF β1可诱导肝细胞凋亡的特性 ,建立体外实验模型 ,观察抗纤方对TGF β1诱导肝细胞凋亡抑制作用。结果 :实验表明TGF β1以剂量依赖关系诱导肝细胞凋亡 ,当TGF β1浓度在 50 0 pg/ml时可诱导2 2 15细胞凋亡达 63% ,而HepG2细胞凋亡只有 4 2 % ;此时 ,用抗纤方 2 0mg/ml可使 2 2 15细胞凋亡率降至 33% ,HepG2细胞凋亡率降至 2 4 % ,抑制率均在 50 %左右。结论 :抗纤方体外对TGF β1诱导的肝细胞凋亡有明显的抑制作用
Objective: To investigate the inhibitory effect of anti-fibrosis on TGFβ1-induced hepatocyte apoptosis. Methods : Using cell biology and other methods, using TGFβ1 can induce the characteristics of hepatocyte apoptosis, establish an in vitro experimental model, observe the inhibitory effect of anti-fibrosis on TGFβ1-induced hepatocyte apoptosis. Results: The experiment showed that TGFβ1 induces hepatocyte apoptosis in a dose-dependent manner. When TGFβ1 concentration is 50 μg/ml, it can induce apoptosis of 225 cells by 63%, while HepG2 cells apoptosis is only 42%. At the time of treatment, the apoptosis rate of the 2215 cells was reduced to 33% with the anti-fibrin 20 mg/ml, and the apoptosis rate of HepG2 cells dropped to 24%, and the inhibition rate was about 50%. Conclusion : Anti-fibrosporin inhibits TGF-β1-induced apoptosis of hepatocytes in vitro
