目的观察β-arrestin2在卵巢切除(OVX)大鼠心脏的表达变化,探索β-arrestin2在雌激素改善OVX大鼠心血管功能中的作用。方法以OVX手术绝经的大鼠为模型,通过监测血中雌激素水平以及子宫质量与体质量比的变化明确OVX模型成功。雌激素替代治疗4周,通过检测心脏质量与体质量比、血压和心率反映心血管功能情况,蛋白质免疫印迹检测心脏β-arrestin1、β-arrestin2和血管紧张素受体(AT1R)蛋白表达水平。结果 OVX组较假手术组血中雌激素水平下降(P<0.05)、子宫萎缩(P<0.05),血压升高(P<0.05),心率加快(P<0.05),心脏体质量比升高(P<0.05);雌激素替代治疗后,血压下降、心率减慢、心脏体质量比降低(P<0.05)。OVX组较假手术组心脏中β-arrestin2表达降低(P<0.05),雌激素可上调其表达,而β-arrestin1和AT1R表达水平无明显变化。结论β-arrestin2在OVX大鼠心脏中表达减少,而雌激素替代治疗可增加β-arrestin2在OVX大鼠心脏组织中的表达。
Objective To investigate the expression of β-arrestin2 in the heart of ovariectomized (OVX) rats and to explore the role of β-arrestin2 in improving the cardiovascular function of OVX rats. Methods OVX surgery in rats as a model of menopause, estrogen levels by monitoring blood and uterine mass to body mass ratio changes OVX model clear success. Estrogen replacement therapy for 4 weeks, cardiac function and body mass ratio, blood pressure and heart rate reflect the cardiovascular function, Western blot detection of cardiac β-arrestin1, β-arrestin2 and angiotensin receptor (AT1R) protein levels. Results The levels of estrogen in OVX group were significantly lower than those in sham group (P <0.05), atrophy in uterus (P <0.05), blood pressure (P <0.05), heart rate (P < (P <0.05). After estrogen replacement therapy, blood pressure decreased, heart rate slowed down, and heart mass ratio decreased (P <0.05). The expression of β-arrestin2 in OVX group was lower than that in sham operation group (P <0.05). The expression of β-arrestin1 and AT1R in esophageal epithelial cells was up-regulated by estrogen. Conclusion β-arrestin2 is decreased in the heart of OVX rats, while estrogen replacement therapy can increase the expression of β-arrestin2 in the heart of OVX rats.
