目的:优化中药碧血胶囊成分配伍方,探讨其对心肌细胞生存率和细胞凋亡的影响。方法:将中药碧血胶囊的4个成分按四因素六水平的均匀设计,对异丙肾上腺素诱导大鼠心肌细胞损伤模型进行了实验干预。采用MTT法检测细胞生存率,流式细胞仪和细胞荧光染色检测心肌细胞凋亡,并通过灰色关联分析其成分配伍方的主次关系。结果:均匀设计得出优化的成分配伍方100μmol/L人参皂苷Rb1+80μmol/L绞股蓝皂苷Ⅳ+15μmol/L荭草苷。灰色关联分析获得该方中各成分的效应关系为绞股蓝皂苷Ⅳ>荭草苷>人参皂苷Rb1。优化的成分配伍方提高细胞生存率作用明显优于单一成分组,其抑制心肌细胞凋亡的效应优于碧血胶囊药物血清(P<0.01)。结论:通过均匀设计与灰色关联分析以及实验验证获得了中药碧血胶囊的优化成分配伍方及其成分效应的主次关系。
OBJECTIVE: To optimize the prescription of traditional Chinese medicine Bi-Xue Capsule and explore its effect on the survival rate and apoptosis of cardiomyocytes. Methods: Four components of Chinese medicine Bi-shen Capsule were designed according to the uniform design of four factors and six levels. Experiments were conducted on isoproterenol-induced cardiomyocyte injury in rats. Cell viability was detected by MTT assay. Cardiomyocyte apoptosis was detected by flow cytometry and cell fluorescence staining. The primary and secondary relationships were also analyzed by gray relational analysis. Results: The optimal design was obtained by uniform design. 100μmol / L ginsenoside Rb1 + 80μmol / L gypenoside Ⅳ + 15μmol / L xyloside. Gray relational analysis of the parties to obtain the effect of each component of the relationship between the gypenoside IV> 荭 glycosides> ginsenoside Rb1. The effect of optimized components compatibility group on cell survival rate was better than that of single component group, and its effect of inhibiting cardiomyocyte apoptosis was better than that of Bi-Xue Capsule (P <0.01). Conclusion: Through the uniform design and gray correlation analysis and experimental verification, we obtained the primary and secondary relationship between the components of the optimized components and the components of the traditional Chinese medicine Bi-Xue Capsules.
