代谢组学是在新陈代谢的动态进程中,系统研究代谢产物的变化规律,揭示生命活动代谢本质的科学。本实验制备正常组和实验组各15只怀孕17.5d的C57BL/6J雌鼠(分别于怀孕10~12d腹腔注射生理盐水和地塞米松)的肝组织水溶性提取液,利用核磁共振技术测定其内源性小分子代谢产物,通过主成分分析方法确定代谢物组成分的变化。实验结果表明,两组样本代谢物组在PCA得分散点图上产生了显著的差异,并且与两组胚胎腭裂发生率的差异相符。所以,采用代谢组学的分析方法可以较好的体现C57BL/6J孕鼠在地塞米松干预下妊娠过程中的代谢物组变化,为唇腭裂致病机理的深入研究开辟了一条新的途径。
Metabolomics is a systematic study of the variation of metabolites in the dynamic process of metabolism to reveal the science of the metabolic essence of life activities. In this study, 15 water-soluble extracts from 15 pregnant 17.5-day-old C57BL / 6J female mice (normal saline and dexamethasone injected intraperitoneally on the 10th to 12th day of gestation) in the normal group and the experimental group were prepared and measured by nuclear magnetic resonance Endogenous small molecule metabolites, by principal component analysis to determine changes in the composition of metabolites. The experimental results show that the two groups of sample metabolites in the PCA score scatter plot produced a significant difference, and with the two groups of embryonic cleft palate the incidence of differences. Therefore, the use of metabonomics analysis can better reflect the changes of metabolites in pregnancy of C57BL / 6J pregnant mice under the intervention of dexamethasone, which opens up a new way for further study on the pathogenesis of cleft lip and palate.
