目的:检测胃癌组织Rac1蛋白表达及其与临床病理特征和生存的关系.方法:运用免疫组化(SP法)结合组织芯片技术联合检测189例胃癌、54例癌旁、30例正常组织及59例淋巴结中Rac1蛋白表达情况.结果:Rac1蛋白在胃癌组织、癌旁组织及正常胃粘膜的表达阳性率分别为72.0%,55.6%,13.3%(P<0.01).Rac1蛋白表达与淋巴结转移、分化程度、浸润深度及lauren分型密切相关(P<0.05).胃癌原发灶Rac1蛋白表达阳性率(82.5%)高于相对应的淋巴结转移灶(60.0%)(P<0.05),差异显著.COX多因素回归分析表明Rac1蛋白表达阳性具有显著的生存危险(P<0.05),是生存期预测因子.结论:Rac1蛋白在胃癌、癌旁组织存在广泛表达,可能是胃癌的早期分子事件;Rac1蛋白表达与临床病理特征密切相关,在胃癌的发生发展及恶性转化过程中起促进作用;Rac1蛋白表达方式与患者的生存期密切相关,其表达可作为独立的生存期预测因子.
OBJECTIVE: To detect the expression of Rac1 protein in gastric carcinoma and its relationship with clinicopathological characteristics and survival.Methods: 189 cases of gastric cancer, 54 cases of paracancerous tissues, 30 cases of normal tissues and 59 normal tissues were detected by immunohistochemistry (SP method) and tissue microarray Rac1 protein expression in lymph node.Results: The positive rates of Rac1 protein expression in gastric cancer tissues, paracancerous tissues and normal gastric mucosa were 72.0%, 55.6% and 13.3%, respectively (P <0.01) .The expression of Rac1 protein and lymph node metastasis, (P <0.05) .The positive rate of Rac1 protein expression in primary gastric cancer (82.5%) was higher than that in corresponding lymph node metastasis (60.0%) (P <0.05), the difference was significant .COX multivariate regression analysis showed that the positive expression of Rac1 protein had a significant risk of survival (P <0.05), which was a predictor of survival.Conclusion: Rac1 protein is widely expressed in gastric cancer and paracancerous tissues and may be an early molecular event in gastric cancer. The expression of Rac1 protein is closely related to clinicopathological features and plays a role in the carcinogenesis and malignant transformation of gastric cancer. The expression of Rac1 protein is closely related to the survival of patients, and its expression may be used as a predictor of survival .
