目的:观察和评价雷公藤内酯醇的半合成衍生物(MC002)对人结肠癌HCT-8小鼠模型的肿瘤抑制作用。方法:用BALB/C裸小鼠接种人结肠癌HCT-8瘤株造成小鼠肿瘤模型,以MC002对荷瘤小鼠的相对肿瘤增殖率、瘤重、抑瘤率为指标评价MC002对小鼠的抑瘤作用。结果:MC002各剂量组瘤体积明显小于模型对照组,瘤重均显著降低,在1.5 mg.kg-1、1.0 mg.kg-1给药10次后T/C%均小于40%(P<0.05),且具有量效关系。结论:MC002能有效抑制人结肠癌HCT-8荷瘤小鼠体内肿瘤生长。通过深入研究MC002有可能发展成为一种新型抗癌药。
Objective: To observe and evaluate the tumor suppressive effect of the semi-synthetic derivative of triptolide (MC002) on human colon cancer HCT-8 mouse model. METHODS: BALB/c nude mice were used to inoculate the human colon cancer HCT-8 tumor strain to create a mouse tumor model. MC002 was used to evaluate the relative tumor proliferation rate, tumor weight and tumor inhibition rate of tumor-bearing mice as an index to evaluate the effect of MC002 on mice. The anti-tumor effect. RESULTS: The tumor volume of each dose group of MC002 was significantly lower than that of the model control group. The tumor weight was significantly decreased. Conclusion: MC002 can effectively inhibit tumor growth in human colon cancer HCT-8 tumor-bearing mice. Through in-depth study of MC002 it is possible to develop a new type of anticancer drug.
