目的研究紫甘薯提取物(purple sweet potato extracts,PSPE)对更年期大鼠生理状态的影响。方法以11~12月龄雌性SD大鼠作为更年期模型,并以3~4月龄雌性大鼠作为中青年对照,对照组喂饲正常饲料,各剂量组大鼠连续饲喂不同剂量的PSPE,8w后观察大鼠的体重变化,计算子宫、卵巢系数,检测血清中雌二醇(E2)、促卵泡激素(FSH)、促黄体激素(LH)含量及子宫组织中雌激素受体(ER)水平。观察PSPE对卵巢组织形态的影响,并采用实时荧光定量PCR法测定子宫ER-α和ER-βm RNA相对表达量。结果与中青年对照组相比,更年期对照组大鼠呈现更年期的改变。而与更年期对照组相比,PSPE可以减缓更年期大鼠体重的持续上升,中剂量PSPE组大鼠的子宫、卵巢系数显著较高,高剂量PSPE组大鼠的血清FSH、LH水平明显较低,PSPE各剂量组大鼠的子宫组织中ER水平较高,卵巢生理学形态得到改善,子宫组织ER-α、ER-βm RNA相对表达量也明显较高。结论 PSPE可以调节大鼠内分泌功能,改善更年期的生理状态。
Objective To study the effects of purple sweet potato extracts (PSPE) on the physiological status of climacteric rats. Methods Female 11-month-old 12-month-old SD rats were used as models for menopause, and 3 to 4-month-old female rats were used as control. The control group was fed normal diet. The rats in each dose group were fed with different doses of PSPE, After 8 weeks, the changes of body weight of the rats were observed, the uterus and ovary coefficients were calculated, the content of estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH) and estrogen receptor (ER) Level. The morphological changes of ovarian tissue were observed by PSPE, and the relative expression of ER-α and ER-βmRNA in uterus was determined by real-time fluorescence quantitative PCR. Results Compared with the young and middle-aged control group, the climacteric control group rats showed climacteric changes. Compared with menopausal control group, PSPE can slow the weight gain of menopause rats continuously. The uterus and ovary of rats in medium dose PSPE group were significantly higher, and the serum FSH and LH levels in high dose PSPE group were significantly lower, The levels of ER in uterine tissue of rats in each dose group of PSPE were higher, the physiological morphology of ovary was improved, and the relative expression of ER-α and ER-βmRNA in uterus was also significantly higher. Conclusion PSPE can regulate the endocrine function of rats and improve the physiological status of menopause.
