目的:观察左归丸及右归丸对老年大鼠学习记忆相关信号转导蛋白表达的影响。方法:以自然衰老(24月龄)SD雄性大鼠为动物模型,随机分为老年对照组、老年皮质酮(GC)拮抗组、老年左归组和老年右归组,另设青年对照组(5月龄)。采用Western Blot法观察各组大鼠学习记忆相关信号转导蛋白表达变化,同时观察补肾方药对上述指标异常变化的改善作用。结果:与青年对照组相比,老年对照组大鼠海马组织学习记忆相关信号分子——CaM、pCaMKⅡ、pERK1/2、pCREB及pSynapsinI、Arc蛋白表达均明显降低(P<0.05);补肾方药左归丸、右归丸能不同程度地改善上述指标的衰老性变化。结论:补肾方药左归丸及右归丸改善老年大鼠空间学习记忆能力的机制之一可能与延缓学习记忆相关信号分子表达的退行性变化有关。
Objective: To observe the effects of Zuogui Pill and Yougui Wan on the expression of learning and memory signal transducers in aged rats. Methods: SD male rats aged 24 months old were randomly divided into three groups: the aged control group, the old antagonist group of corticosterone (GC), the Zuogui group of the aged and the Yougui group of the aged group, and another young control group ( 5 months old). Western Blot method was used to observe the changes of learning and memory signal transduction proteins in each group, meanwhile, to observe the effect of Bushen Recipe on the abnormal changes of the above indexes. Results: Compared with the young control group, the expressions of CaM, pCaMKⅡ, pERK1 / 2, pCREB, pSynapsin and Arc in the hippocampus of the aged control group were significantly decreased (P <0.05) Gui Wan, You Gui Wan can improve the above indicators of varying degrees of aging changes. Conclusion: One of the mechanisms of Bushen Recipe Zuogui Pill and Yougui Pill to improve spatial learning and memory in aged rats may be related to delaying the degenerative changes of signal molecules involved in learning and memory.
