目的:建立新生儿血斑中未衍生化游离氨基酸的串联质谱检测方法,为氨基酸代谢性疾病的诊断提供新的思路。方法:应用超高效液相色谱-三级四极杆串联质谱,采用多反应监测(MRM)扫描模式同时结合产物离子确证扫描(PICS),对新生儿血斑未衍生化游离氨基酸进行定性定量分析。结果:在1 min内可以同时定量分析9种氨基酸,分别为Phe、Tyr、Pro、Val、Thr、Leu(XIe)、Lys(Gln)、Met、Glu,而且每种氨基酸在0.5~200μmol/L范围内均有良好的线性关系。结论:PICS模式可以在真正的血斑样品分析中,提供更加准确的一种或几种氨基酸的定性信息,建立的方法适合于新生儿血斑样本的临床分析及研究。
OBJECTIVE: To establish a tandem mass spectrometry method for the determination of non-derivatized free amino acids in newborn blood spots, and provide a new idea for the diagnosis of amino acid metabolic diseases. Methods: Qualitative and quantitative analysis of underivatized free amino acids in neonates was performed by ultra performance liquid chromatography-triple quadrupole tandem mass spectrometry (MS-MS / MS) coupled with product ion confirmation scanning (MRS) . Results: Nine amino acids were quantitatively analyzed in 1 min, which were Phe, Tyr, Pro, Val, Thr, Leu, Lys, Met and Glu, The range has a good linear relationship. Conclusion: The PICS model can provide more accurate qualitative information of one or more amino acids in real blood spot sample analysis. The established method is suitable for the clinical analysis and research of neonatal blood spot samples.
