目的:通过大鼠胚胎发育期予补肾方干预,以研究中医补肾方在胚胎发育期干预能否预防高脂饮食诱发的子代成年大鼠糖耐量低减(IGT)及其对胰岛素、瘦素的影响。方法:将孕鼠分为正常对照组、模型组、左归丸组、右归丸组、八珍汤组,在孕1~19 d,各用药组分别以左归丸、右归丸、八珍汤灌胃(ig,20 g·kg-1,20 mL·kg-1),正常对照组和模型组以生理盐水灌胃。除正常对照组外,其余各组子鼠均于6周龄开始高脂饲料喂养,12周后测各组子鼠空腹血糖(FBG)、糖负荷后2 h血糖(2hBG)、血清空腹胰岛素、瘦素。结果:与模型组比较,左归丸组2hBG明显降低(P<0.05);八珍汤组和右归丸组2hBG无明显降低。左归丸组、右归丸组、八珍汤组胰岛素抵抗指数(HOMA-IR)均较模型组显著降低(P<0.01,P<0.01,P<0.05)。左归丸组和右归丸组血清瘦素显著低于模型组(P<0.01,P<0.01);而八珍汤组血清瘦素水平与模型组相比无显著性差异。结论:左归丸与右归丸胚胎期干预都可对抗高脂饮食引发子代成年大鼠胰岛素抵抗、瘦素抵抗,但对IGT的预防作用以左归丸为优。八珍汤组对抗胰岛素抵抗、瘦素抵抗作用相对弱,也未能预防高脂饮食引起的子鼠IGT的发生。
OBJECTIVE: To investigate the effect of Bushen Decoction during embryonic development on prevention of impaired glucose tolerance (IGT) in adult rats induced by high-fat diet and its effect on insulin, leptin Impact. Methods: Pregnant rats were divided into normal control group, model group, Zuoguiwan group, Youguiwan group and Bazhen decoction group. The pregnant rats were randomly divided into two groups: Zuogui pill, Yougui pill, Zhenpeng gavage (ig, 20 g · kg-1, 20 mL · kg-1), the normal control group and model group by normal saline gavage. Except for the normal control group, the other groups were fed with high-fat diet at 6 weeks of age. After 12 weeks, fasting blood glucose (FBG), fasting blood glucose (2hBG), serum fasting insulin Leptin. Results: Compared with the model group, the 2hBG in Zuoguiwan group was significantly decreased (P <0.05); the BZG group and Yougui Wan group had no significant decrease in 2hBG. The insulin resistance index (HOMA-IR) in Zuoguiwan group, Youguiwan group and Bazhen decoction group were significantly lower than those in model group (P <0.01, P <0.01, P <0.05). Serum leptin concentrations in Zuoguiwan and Yougui Wan groups were significantly lower than those in model group (P <0.01, P <0.01), while serum leptin levels in Bazhen decoction group were not significantly different from those in model group. Conclusion: Both Zuogui Pill and Yougui pill embryos can resist insulin resistance and leptin resistance in offspring of offspring against high-fat diet, but Zuogui pill is superior to IGT. Bazhen decoction group has a relatively weak effect on insulin resistance and leptin resistance, and it also fails to prevent the occurrence of IGT in the offspring induced by a high-fat diet.
