[目的]检测结肠癌组织中趋化因子受体1(CXCR1)基因表达的改变,探讨其相关的临床意义。[方法]选取2014年3月~2016年6月我院收治的结肠癌患者手术切除癌组织86例,以及相应的癌旁正常结肠黏膜组织(距离肿块组织≥5cm)。SYBR Green荧光定量-PCR检测CXCR1基因表达量。[结果]结肠癌肿瘤组织中CXCR1基因表达量为0.407±0.064,显著高于对照非肿瘤肠黏膜组织的0.215±0.037(P<0.01)。结肠癌肿瘤组织中CXCR1基因表达量在男女性别和不同年龄间差异无统计学意义(P>0.05)。有淋巴结转移肿瘤组织CXCR1基因表达量为0.458±0.073,显著高于无淋巴结转移肿瘤组织的0.346±0.051(P<0.05)。中、低分化肿瘤组织CXCR1基因表达量为0.432±0.071,显著高于高分化肿瘤组织的0.358±0.056(P<0.05)。Ⅲ、Ⅳ期肿瘤组织CXCR1基因表达量为0.462±0.078,显著高于Ⅰ、Ⅱ期肿瘤组织的0.365±0.054(P<0.05)。[结论]CXCR1基因在结肠癌组织中表达上调,CXCR1基因表达上调与结肠癌有无淋巴结转移、分化程度以及临床分期密切相关。
[Objective] To investigate the change of chemokine receptor 1 (CXCR1) gene expression in colon cancer and to explore its clinical significance. [Methods] From March 2014 to June 2016, 86 cases of resected cancerous tissue from patients with colon cancer admitted to our hospital and corresponding adjacent normal colonic mucosa tissues (≥5cm from tumor mass) were selected. SYBR Green Fluorescence Quantitative-PCR Detection of CXCR1 Gene Expression. [Results] The expression of CXCR1 gene in colon cancer tissue was 0.407 ± 0.064, which was significantly higher than that of the control non-tumor intestinal mucosa (0.215 ± 0.037, P <0.01). There was no significant difference in the expression of CXCR1 gene in colon cancer tissues between male and female gender and age (P> 0.05). The expression of CXCR1 in lymph node metastasis was 0.458 ± 0.073, which was significantly higher than that in non-lymph node metastasis (0.346 ± 0.051, P <0.05). The expression of CXCR1 in moderate and poorly differentiated tumors was 0.432 ± 0.071, which was significantly higher than that in well differentiated tumors (0.358 ± 0.056, P <0.05). The expression of CXCR1 gene in stage Ⅲ and Ⅳ tumors was 0.462 ± 0.078, which was significantly higher than that in stage Ⅰ and Ⅱ tumors (0.365 ± 0.054, P <0.05). [Conclusion] The expression of CXCR1 gene is up-regulated in colon cancer tissues. The upregulation of CXCR1 gene expression is closely related to the presence or absence of lymph node metastasis, differentiation and clinical stage of colon cancer.
