以核桃(Juglans regia)的不同部位(根、1年生茎、2年生茎和干茎)为材料,采用组织分离法,对来自陕西山阳县、蓝田县与宜君县3个不同生境的材料进行内生真菌的分离鉴定、种群组成、定殖率、多样性指数等研究。结果显示,从576块组织块中共分离得到392株内生真菌,其中在2年生茎124株,1年生茎120株,干茎87株,根61株。392株内生真菌隶属于24个属,其中,3大优势类群为交链孢霉属(Alternaria)、花核菌属(Anthina)和茎叶核菌属(Ectostroma),分别占菌株总数的19.90%、9.69%和8.93%。交链孢霉属主要存在于2年生茎和干茎中,花核菌属和茎叶核菌属则主要存在于2年生茎中。核桃不同部位内生真菌多样性指数1年生茎的最高,为1.07;不同生境中,山阳核桃内生真菌的多样性指数最高,为1.30,说明春季核桃内生真菌具有丰富的多样性,是潜在的真菌资源库。
Taking the different parts of the Juglans regia (root, 1-year-old stem, 2-year-old stem and stem) as materials, the tissue isolation method was used to study the effects of the materials from 3 different habitats in Shanyang County, Lantian County and Yijun County, Endophytic fungi were isolated and identified, population composition, colonization rate, diversity index and other research. The results showed that 392 endophytic fungi were isolated from 576 tissue pieces, of which 124 were on 2-year-old stems, 120 on 1-year-old stems, 87 on dried stems and 61 on roots. 392 strains of endophytic fungi belonging to 24 genera, among them, the three dominant groups were Alternaria, Anthina and Ectostroma, accounting for 19.90 %, 9.69% and 8.93% respectively. The species of Alternaria is mainly found in 2-year-old stems and stems, while the genus Borrelia and Sphaerotheca are mainly found in 2-year-old stems. In different habitats, the highest endophytic fungi diversity index of endophytic fungi was 1.07. The diversity index of endophytic fungi in Sanyang walnut was the highest at 1.30 in different habitats, which indicated that endophyte was rich in diversity in spring and Potential fungal resource base.
