目的 了解北京地区 1990 - 1991和 1997- 1998两个非连续的流行年中呼吸道合胞病毒 (RSV)A、B亚型和基因型的流行情况。方法 用间接免疫荧光法 (IIF)检测呼吸道合胞病毒阳性鼻咽分泌物 (NPS)标本或RSV分离株 ,划分A、B亚型。根据N基因片段的限制性酶切图型将RSV分离株分成至少 6个基因型NP1- 6 ,其中NP1、3和 6属于B亚型 ,NP2、4和 5属于A亚型。根据SH基因片段的核苷酸序列将A亚型分离株进一步划分为SH基因型SHL1- 6。SHL之间关系密切 ,并与NP有关。SHL1、3和 4紧密相关 ,属于NP2 ;SHL2和 6属于NP4 ;SHL5属于NP5。结果 1997冬季至 1998年春季 145份RSVNPS标本中 ,83份 (57.2 % )为B亚型 ,6 2份 (4 2 .8% )为A亚型 ,A :B为 1:1.3。 1997- 1998流行年所获的 10株RSV分离株中 ,2株为A亚型 ,8株为B亚型 ,A :B为 1:4。 1990 - 1991流行年所获的 10株毒株中 ,8株为A亚型 ,2株为B亚型 ,A :B为 4 :1。 1997- 1998流行年分离到的 10株RSV分离株中 ,2株A亚型均属NP4 ,SHL2 ;8株B亚型均为NP3。 1990 - 1991流行年分离得到的 10株毒株中 8株A亚型均属NP4 ,SHL2 ;而 2株B亚型均为NP3。根据NP推导出的A、B亚型划分与单克隆抗体定义的亚型一致。结论 北京地区RSVA、B亚型或多个基因型可同时流行 ,但每个流行年的优
Objective To understand the prevalence of respiratory syncytial virus (RSV) subtypes A, B and genotypes in two non-consecutive epidemic years of 1990-1991 and 1997-1998 in Beijing. Methods Respiratory syncytial virus positive nasopharyngeal secretions (NPS) or RSV isolates were detected by indirect immunofluorescence (IIF), and subtype A and B were divided into two groups. The RSV isolates were divided into at least 6 genotypes, NP1-6, based on the restriction map of the N gene fragment, with NPs 1, 3 and 6 belonging to the B subtype and NP2, 4 and 5 belonging to the A subtype. According to the nucleotide sequence of the SH gene fragment, the subtype A subtype was further divided into the SH genotype SHL1-6. SHL is closely related, and with the NP. SHL1, 3 and 4 are closely related and belong to NP2; SHL2 and 6 belong to NP4; SHL5 belong to NP5. Results Of the 145 RSVNPS specimens from the winter of 1997 to the spring of 1998, 83 (57.2%) were subtypes B, 62 (42.8%) were subtypes A, and A: B was 1: 1.3. Of the 10 RSV isolates obtained during the 1997/1998 epidemic year, 2 were subtype A, 8 were subtype B, and A: B was 1: 4. Of the 10 strains collected in the 1990-1991 epidemic year, 8 were subtype A, 2 were subtype B, and A: B was 4: 1. Of the 10 RSV isolates isolated from the 1997/1998 epidemic year, 2 were subtype A and 4 were NP4 and SHL2, respectively. All 8 subtypes were NP3. Among the ten isolates isolated from 1990 to 1991 in the epidemic year, all the 8 A subtypes belonged to NP4 and SHL2, while the two subtypes B were NP3. According to the NP derived A, B subtype and monoclonal antibody defined subtype. Conclusions RSV A, B genotypes or multiple genotypes in Beijing area may be prevailing at the same time, but the advantages of each popular year
