野生干巴菌(Thelephora ganbajun)产量有限,探索其人工或半人工栽培方法自然成为人们关注的问题。而干巴菌菌种的分离则是研究其生活史及人工栽培的前提条件之一,对干巴菌繁殖具有重要意义。本工作在于探索分离干巴菌菌种的合适方法。 1 试验材料 1.1 子实体 采集于昆明小哨的成熟新鲜子实体(灰白或褐色,帚状小分枝明显,顶端小枝片达5cm以上)和幼嫩子实体(鲜黄色,未见珊瑚状分枝,直径0.5~2cm)。 1.2 培养基 斜面培养基为马铃薯20g,葡萄糖20g,硫酸镁0.5g,磷酸二氢钾0.25g,硫胺素30μg,琼脂20g,水1000mL,121℃灭菌30分钟制成斜面。A培养基:将上述灭菌培养基熔化,稍冷却后在无菌室内加链霉素50μg/mL,摇匀,倒入灭菌培养皿,每皿1O~15mL,厚
