目的:探讨外周血内皮祖细胞(EPCs)数量及动脉内皮细胞型一氧化氮合酶(eNOS)基因表达在动脉粥样硬化(AS)大鼠中的改变。方法:将70只大鼠分为3组:正常对照组20只,采用假手术+普通饲料喂养;实验对照组20只,采用假手术+高脂饲料喂养;AS组30只,采用球囊损伤主动脉内皮、维生素D33×105 U·kg-1·d-1分6次肌内注射和饲以高脂饲料喂养。90d后分别采外周血进行EPCs的分离培养,于倒置相差显微镜下计数内皮祖细胞克隆形成单位(EPC-CFU),并采用Real-time PCR法检测腹主动脉中eNOS基因的表达。结果:与正常对照组和实验对照组相比,AS组外周血EPCs数量明显减少(P<0.01),eNOS基因表达明显下调(P<0.01);EPCs数量及eNOS表达在正常对照组和实验对照组中差异无统计学意义。结论:AS时EPCs数量减少,eNOS基因表达明显下调。
Objective: To investigate the changes of the number of endothelial progenitor cells (EPCs) and the expression of endothelial nitric oxide synthase (eNOS) gene in atherosclerosis rats. Methods: Totally 70 rats were divided into 3 groups: normal control group (n = 20) and sham operation + normal diet group (n = 20). The rats in experimental group were fed with sham operation and high fat diet. Aortic endothelial, vitamin D33 × 105 U · kg-1 · d-1 6 intramuscular injection and feeding high-fat diet. After 90 days, EPCs were isolated and cultured in peripheral blood. The EPC-CFU of endothelial progenitor cells was counted under inverted phase contrast microscope. The expression of eNOS gene in abdominal aorta was detected by Real-time PCR. Results: Compared with normal control group and experimental control group, the number of EPCs in AS group was significantly decreased (P <0.01) and eNOS gene expression was significantly decreased (P <0.01). The number of EPCs and eNOS expression in normal control group and experimental control group There was no significant difference in the group. Conclusion: The number of EPCs in AS decreased and the expression of eNOS gene was significantly down-regulated.
