目的:分析大鼠灌胃白花蛇舌草有效部位提取物后血浆中对香豆酸、反式6-O-对香豆酰鸡屎藤苷甲酯及其代谢产物。方法:取健康雄性SD大鼠按0.4 g.kg-1的剂量经口给予白花蛇舌草有效部位提取物,1.0 h后采集眼眶静脉血液制备血浆,采用HPLC-Q-TOF-MS进行分析。结果:在大鼠血浆中只检测到1个代谢产物,并利用对照品鉴定了其结构为对香豆酸。结论:白花蛇舌草有效部位提取物中主要成分反式6-O-对香豆酰鸡屎藤苷甲酯在大鼠体内的代谢产物为对香豆酸,本实验为研究白花蛇舌草化学成分及药效作用提供了理论依据。
Objective: To analyze the plasma p-coumaric acid, trans-6-O-p-coumaroyl fetidin methyl ester and its metabolites in rats after intragastric administration of the extract of Hedyotis diffusa Willd. Methods: Healthy male Sprague-Dawley rats were orally administered with 0.4 g.kg-1 dose of extract of Hedyotis diffusa. 1.0 h later, orbital venous blood was collected for plasma preparation and analyzed by HPLC-Q-TOF-MS. Results: Only one metabolite was detected in the plasma of rats and its structure was identified as p-coumaric acid. CONCLUSION: The metabolite of trans-6-O-coumarate chicken fematidine methyl ester in the active fraction of the Hedyotis diffusa will be p-coumaric acid. In this study, Chemical composition and pharmacodynamic effects provide a theoretical basis.
