目的观察补脏通络方对糖尿病合并大血管病变大鼠主动脉内膜MMP-9/TIMP-1表达的影响,探讨其逆转糖尿病大血管内膜增厚的可能机制。方法脂肪乳灌胃21d后一次性尾静脉注射STZ 30mg/kg造模。补脏通络方治疗。病理切片光镜观察大鼠胸主动脉内膜形态改变,检测内膜厚度;免疫组化检测主动脉内膜IV型胶原表达;免疫印迹法分析主动脉内膜MMP-9、TIMP-1蛋白表达。结果与正常组相比,病理组主动脉内膜增厚明显(P<0.01),IV型胶原表达增加(P<0.01),MMP-9、TIMP-1表达增加(P<0.01),MMP-9/TIMP-1比值上升(P<0.01);经补脏通络方治疗,与病理组相比,各治疗组主动脉内膜厚度和IV型胶原表达降低(P<0.01),MMP-9表达降低(P<0.01),TIMP-1表达增加(P<0.01),MMP-9/TIMP-1比值下降(P<0.01);补脏通络方高剂量组降低MMP-9、升高TIMP-1的作用优于低剂量组(P<0.05)。结论调节大血管内膜MMP-9/TIMP-1平衡,加速胶原降解,可能是补脏通络方逆转糖尿病大血管内膜增厚的可能机制。
Objective To observe the effect of “Bu Dang Tongluo Prescription” on the expression of MMP-9 / TIMP-1 in the intima of diabetic rats with macroangiopancosis and to explore its possible mechanism of reversing the intimal thickening in diabetic rats. Methods The fat emulsion was injected into STZ 30mg / kg after 21 days via intragastric administration. Dirty Tongluo side treatment. The pathological changes of thoracic aorta were observed by light microscopy. The thickness of intima was detected by immunohistochemistry. The expression of type IV collagen in the aorta was detected by immunohistochemistry. The expressions of MMP-9 and TIMP-1 in the aortic intima were detected by immunoblotting . Results Compared with the normal group, the aortic intima thickening was significant (P <0.01), the type IV collagen expression was increased (P <0.01), the expression of MMP-9 and TIMP-1 was increased 9 / TIMP-1 ratio increased (P <0.01) .Compared with pathological group, compared with pathological group, the aortic intima thickness and the expression of type IV collagen decreased (P <0.01) (P <0.01), and the ratio of MMP-9 / TIMP-1 was decreased (P <0.01). Compared with the other two groups, the expression of TIMP-1 was down-regulated -1 is better than low dose group (P <0.05). Conclusions The regulation of the balance of MMP-9 / TIMP-1 in the intima of large vessels and the acceleration of collagen degradation may be the possible mechanism of Bu Dang Tong Luo Recipe in reversing the intimal thickening of diabetic vessels.
