目的对2011年广州市分离的6株脑膜炎奈瑟菌(Neisseria meningitidis,N.meningitidis)进行分子特征分析。方法提取菌株基因组DNA,通过DNA序列测定分析外膜蛋白(outer membranes proteins,OMPs)编码基因porA、fetA可变区型别;通过多位点序列分型(multi-locus sequence typing,MLST),采用Splits Tree在线软件分析进化关系,研究其分子特征。结果 6株菌株的OMPs编码基因porA、fetA可变区型别均不相同;MLST分型显示其分属5个不同的序列型(Sequence Type,ST),其中2株B群菌株序列型为ST-5615,1株C群菌株序列型为ST-5542,1株血清不能分群菌株序列型为ST-2146,分离自健康带菌者和密切接触者的2株W135群菌株的序列型不同,其中密切接触者为高致病性的ST-11型,而健康带菌者为新发现的ST-9133型。结论广州市N.meningitidis的基因型具有多样性特点,本研究为完善我市N.meningitidis分型数据、研发新疫苗及免疫规划措施的制订提供参考。
Objective To analyze the molecular characteristics of Neisseria meningitidis (N. meningitidis) isolated in Guangzhou in 2011. Methods The genomic DNA of the strains was extracted and the variable regions of porA and fetA encoding genes of outer membranes proteins (OMPs) were analyzed by DNA sequencing. By multi-locus sequence typing (MLST) Splits Tree online software analyzes evolutionary relationships and studies their molecular characteristics. Results The polymorphisms of porA and fetA in OMPs of 6 isolates were different. MLST typing showed that they belong to 5 different sequence types (ST) -5615, 1 strain C group Strain ST-5542, 1 strain can not be stratified Strain ST-2146, isolated from healthy carriers and close contacts of two strains of W135 different sequence type, in which close The contacts were of the highly pathogenic ST-11 type, while healthy carriers were the newly discovered type ST-9133. Conclusion The genotypes of N. meningitidis in Guangzhou are characterized by diversity. This study provides a reference for improving the typing data of N. meningitidis and developing new vaccines and immunization planning measures in our city.
