脂质体平衡透析与液质联用技术筛选脊痛宁片物质基础研究

目的研究脊痛宁片有效成分与模拟生物膜的相互作用,预测药物在体内的吸收情况。方法将脂质体作为模拟生物膜,采用脂质体平衡透析与液质联用技术筛选脊痛宁片中与脂质体有相互作用的成分,并考察脂质体质量浓度和缓冲液p H值对脊痛宁片与脂质体互相作用的影响。结果脊痛宁片中共有11个成分与脂质体作用明显,包括4个黄酮类成分,2个萜类成分,4个香豆素类成分,1个苯丙酸类成分。脂质体的质量浓度和缓冲液p H值都对脊痛宁片与脂质体互相作用有一定的影响。结论脂质体平衡透析与液质联用可预测脊痛宁片潜在的活性成分在体内的吸收情况,为其药效物质基础的进一步研究提供依据。 Objective To study the interaction between the effective components of Jiutongning tablet and simulated biofilm and predict the absorption of drugs in the body. Methods The liposomes were used as simulated biofilms. Liposomes were used to screen the components that interact with liposomes by liposome balance dialysis and liquid chromatography-mass spectrometry (LC-MS / MS), and the liposomes were separated from pH Effect of Jiaotongning Tablet and Liposome on the Interaction of. Results There were 11 components in Jiutongning tablet which had obvious effects on liposomes, including 4 flavonoids, 2 terpenoids, 4 coumarins and 1 phenylpropanoid. Both the concentration of liposomes and the p H value of buffer had a certain influence on the interaction between Jiitongning tablets and liposomes. Conclusion Liposomal equilibrium dialysis and liquid chromatography-mass spectrometry (LC-MS / MS) can predict the absorption of potential active ingredients in Jiutongning tablets and provide a basis for further study of its pharmacodynamic substance.
