目的 探讨c erbB2和p5 3基因扩增和表达在口腔鳞癌 (OSCC)发生发展中的作用。 方法 用原位杂交和免疫组织化学技术与图像分析相结合的方法 ,对 30例口腔鳞癌和 5例正常口腔粘膜中c erbB2和p5 3两种基因的扩增、mRNA表达和蛋白表达状况进行了定性、定位、定量研究。 结果 OSCC中c erbB2和p5 3的扩增阳性率分别为 4 6 6 7%和 4 0 % ;而c erbB2和p5 3在OSCC中的mRNA表达率分别是 70 %和 80 % ,蛋白表达率分别是4 6 6 7%和 6 3 33%。这两种基因的扩增、mRNA表达和p5 3蛋白表达在各种性状差异之间均没有显著性差异 (P >0 0 5 ) ;erbB2蛋白表达在不同部位的口腔鳞癌中具有显著性差异 (P <0 0 2 )。 结论 c erbB2和p5 3两种基因的扩增和过表达在OSCC发生发展中都具有重要作用 ,c erbB2蛋白表达增高在腭癌和口底癌的形成过程中具有更重要的意义。
Objective To investigate the roles of cbbB2 and p5 3 gene in the development of oral squamous cell carcinoma (OSCC). Methods The combination of in situ hybridization, immunohistochemistry and image analysis was used to amplify the mRNA and protein expressions of c erbB2 and p5 3 in 30 cases of oral squamous cell carcinoma and 5 cases of normal oral mucosa. The qualitative, positioning, quantitative research. Results The positive rates of c-erbB2 and p5 3 in OSCC were 46.67% and 40.0%, respectively. The mRNA expression rates of c-erbB2 and p-53 in OSCC were 70% and 80%, respectively Is 46 6 7% and 6 3 33%. There was no significant difference in the amplification, mRNA expression and p5 3 protein expression between these two genes (P> 0.05). The expression of erbB2 protein in oral squamous cell carcinoma at different sites showed significant difference (P <0 0 2). Conclusion The amplification and overexpression of two genes c erbB2 and p5 3 play an important role in the development of OSCC. The increase of c erbB2 protein expression is more important in the formation of palatal carcinoma and oral floor carcinoma.
