目的 探讨Oncolyn对石英粉尘致肺泡巨噬细胞 (AM )氧化损伤的缓解作用。方法 采用肺灌洗法获得AM后与石英粉尘及Oncolyn共同培养 18h后 ,用单细胞凝胶电泳检测其DNA损伤状况 ,同时检测AM抗氧化酶超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH Px)及一氧化氮合酶 (NOS)的活性 ,以及培养液中一氧化氮 (NO)的含量。结果 大鼠AM与石英粉尘共同培养后 ,彗星出现率、DNA迁移长度增加 ,NO、NOS水平上升 ,SOD、GSH Px的活性下降。在培养液中加入不同浓度的Oncolyn后 ,AM中彗星出现率、DNA迁移长度、NO、NOS的水平均有不同程度的下降 ,且还能提高AM中SOD、GSH Px的活性。结论 Oncolyn对石英粉尘所致某些生化指标有一定的缓解作用
Objective To investigate the relieving effect of Oncolyn on oxidative damage of alveolar macrophages (AM) induced by quartz dust. Methods After the lung lavage method was used to get the AM, it was co-cultured with quartz dust and Oncolyn for 18 hours. The DNA damage status was detected by single cell gel electrophoresis. The activities of superoxide dismutase (AM), glutathione Peroxidase (GSH Px) and nitric oxide synthase (NOS), as well as the content of nitric oxide (NO) in culture medium. Results After co-cultured AM and quartz dust, the incidence of comet and the length of DNA migration increased, the levels of NO and NOS increased and the activities of SOD and GSH Px decreased. The addition of different concentrations of Oncolyn to the culture medium resulted in the decrease of the occurrence rate of comet, the length of DNA migration, the levels of NO and NOS in AM and the activities of SOD and GSH Px in AM. Conclusion Oncolyn can relieve certain biochemical parameters caused by quartz dust
